General description
Lipoxygenases (LOX) are dioxygenases that do not contain heme iron. These enzymes convert unsaturated fatty acids into epoxides; for example, the synthesis of leukotrienes from arachidonic acid. They play an important role in cell proliferation, differentiation, and inflammation; and they have been implicated in diseases mediated by inflammation and hyperproliferation such as asthma, rheumatoid arthritis, and cancer.
Suitability
Suitable for the measurement of lipoxygenase activity in cell and tissue lysates, biological fluids (eg serum), recombinant enzyme, and purified protein.
Beginning
In the lipoxygenase activity assay kit, lipoxygenase converts the LOX substrate to an intermediate that reacts with the probe to generate a fluorescent product. The increase in fluorescent signal can be recorded at λex = 500 nm / λem = 536 nm and is directly proportional to the LOX activity. The kit includes the enzyme 5-lipoxygenase (LOX enzyme) as a positive control. Also included is a lipoxygenase inhibitor that completely inhibits lipoxygenase activity to calculate specific LOX activity in biological samples.
